Application

o-Phenylendiamin wird verwendet, um Transforming Growth Factor (TGF-β) nachzuweisen und Anti-cTfRMAb-ScFv-Fusionsprotein-Antikörper mittels Enzyme-linked Immunosorbent Assay (ELISA) zu messen.

o-Phenylendiamin (freie Base) ist ein Peroxidase-Substrat zur Verwendung in ELISA-Verfahren. Das Substrat produziert ein lösliches Endprodukt mit orange-brauner Färbung, das bei 450 nm spektrophotometrisch abgelesen werden kann. Die OPD-Reaktion kann mit 3 N HCl oder 3 M H₂SO₄ gestoppt und bei 492 nm abgelesen werden.

Biochem/physiol Actions

o-Phenylendiamin (o-PD) ist ein wirksames Substrat für den spektrophotometrischen Meerrettichperoxidase(HRP)-vermittelten Enzyme-linked Immunosorbent Assay (ELISA), der 2,3-Diaminophenazin (DAP) als ein Produkt der o-PD-Oxidation erzeugt.

Preparation Note

Lösen Sie eine Tablette in 0,05 M Phosphat-Citrat-Puffer, pH-Wert 5,0, um die gewünschte Konzentration zu erhalten (in der Regel wird eine OPD-Konzentration von 0,4 mg/ml verwendet). Geben Sie unmittelbar vor der Verwendung 40 μl frisches 30%iges Wasserstoffperoxid je 100 ml Substratpufferlösung hinzu. Die Tabletten sind einzeln in Folienpackungen verpackt.
