生物化学的／生理学的作用
ホタルルシフェラーゼは、発光を触媒する62 kDaのタンパク質です。本酵素は、2段階のプロセスで発光させるために、ATP、分子状酸素および複素環化合物であるホタルルシフェリンを必要とします。発光反応はルシフェリン活性化（そのカルボキシレート基のアデニル化）により開始され、分子状酸素の存在下で進行して黄緑色光の光子を生成します。
単位の定義
ルシフェラーゼの1酵素単位は、20～25°Cで10秒間に1相対光単位（RLU）が生成し、これはGloMax 20/20照度計（ルミノメーター）を使用してTris-グリシン緩衝液（pH 7.6）中に40 pmolのATPと15 nmolのルシフェリンを含有する100 μLのアッセイ混合液中で測定されます。

物理的形状
HEPES（pH 7.5）、NaCl、MgCl2、EDTA、DTTおよび炭水化物安定剤を含有する凍結乾燥粉末として提供されます。
この製品は、E. coli（大腸菌）のluc遺伝子発現で産生されたPhotinus pyralis（アメリカホタル）の組換えルシフェラーゼ（62 kDa）です。
調製ノート
最大の溶解度を得るためには、カウンターイオンを含む1 M Tris緩衝液などpH 7～8の高い塩濃度で酵素を復元調製することが重要です。酵素は、最大5 mgタンパク質/mLの濃度で調製することができます。ボルテックスや攪拌はしないでください。
復元調製後、酵素溶液は4～8°Cで最大2日間維持するか、ワーキングアリコートにして-20°Cで少なくとも1ヵ月間凍結できます。凍結融解を繰り返すことはお勧めしません。
