A1120はレチノール結合タンパク質4(RBP4)に選択的な非レチノイド性のリガンドです。RBP4は肝臓から肝臓外の組織へレチノールを輸送し、RBP4の低下はマウスのインスリン感受性を改善すると報告されています。A1120はRBP4に対する高親和性の非レチノイド性リガンドであり、RBP4とその結合パートナーであるトランスサイレチン (TTR)の相互作用を妨害します。レチノールと同じ部位に結合し、RBP4-TTR相互作用の接触面で(レチノール結合型と比較して)ループの配向に変化を引き起こします。A1120は、RBP4とレチノールのレベルを用量依存的にフェンレチニドの場合と同程度に低下させます。しかしフェンレチニド(Sigma H7779)とは異なり、A1120はインスリン抵抗性に対して有効な作用を示しません。
