Allgemeine Beschreibung
Temozolomid ist ein kleines lipophiles Alkylans.[1] Es interagiert kovalent mit der nukleophilen Mikroumgebung in der DNA (Guaninreste). Aufgrund dieser Eigenschaft gilt Temozolomid als zytotoxischer Wirkstoff. Der Wirkmechanismus von TMZ hängt von seiner Fähigkeit ab, DNA zu hydrolysieren, was zu einem DNA-Abbau und zur Zerstörung von Tumorzellen führt. Maligne Gliomzellen reagieren auf TMZ primär durch G2/M-Zellzyklusarrest oder in seltenen Fällen durch Apoptose.[1] Bei Temozolomid handelt es sich um ein DNA-Methylierungsmittel sowie ein die Arzneimittelresistenz modifizierendes Mittel mit Anti-Tumor- und anti-angiogenen Eigenschaften. Temozolomid löst eine G2/M-Hemmung und Apoptose durch Adduktion einer Methylgruppe an die O6-Position von Guanin in genomischer DNA und durch funktionelle Inaktivierung des DNA-Reparaturproteins O(6)-Alkylguanin-DNA-Alkyltransferase (AGT) im Pfad der Basen-Exzisionsreparatur (BER) aus.
Anwendung
Temozolomid wird zur Analyse von Mechanismen der Arzneimittelresistenz bei Glioblastom-Zelllinien verwendet. Temozolomid wird zur Induktion zytotoxischer Wirkungen auf Glioblastom-Zellen verwendet, um das Ergebnis der Inhibition von Proteindisulfid-Isomerase (PDI) zu untersuchen.[2]

How long is Temozolomide stable for when dissolved as recommended, in DMSO at 10 mg/mL and storaged at 2-8C? Also, can the solution only be stored at 2-8C or can it be stored at -20C or -80C for long term storage

Wherever possible, solutions should be prepared fresh and used on the same day. However, should stock solutions need to be prepared in advance, store in aliquots at -20°C for up to 1 month. Allow aliquots to equilibrate to room temperature prior to opening. The long term storage at other temperatures is not determined. However, the published reference below may prove helpful:
