General description
Humane α-Amylase besteht aus 496 Aminosäuren in einer einzelnen Polypeptidkette, die auf Chromosom 1 kodiert ist. Es wird entweder in der Schilddrüse oder der Pankreas produziert. Die Speichel- und Pankreas-α-Amylasen haben eine homologe Primärsequenz, weisen aber unterschiedliche Spaltungsmuster auf.[1]

Application
α-Amylase wird zur Hydrolyse von α-Bindungen α-verknüpfter Polysaccharide wie Stärke und Glykogen verwendet. Produkt A1031 stammt aus menschlichem Speichel, ist vom Typ IIA und wird als lyophilisiertes Pulver geliefert. α-Amylase wird in verschiedenen Pflanzenstudien wie beispielsweise zur Untersuchung des Stoffwechsels in Arabidopsis eingesetzt[2]. α-Amylase aus menschlichem Speichel wird zur Untersuchung der Entwicklung von Nutrazeutika verwendet, die in der Behandlung von Diabetes und Adipositas hilfreich sein können .
Biochem/physiol Actions
α-Amylase hydrolysiert die α-(1,4)-Glucanverknüpfungen in Polysacchariden von drei oder mehr α-(1,4)-verknüpften D-Glucoseeinheiten. Natürliche Substrate wie Stärke oder Glykogen werden in Glucose und Maltose zerlegt.[3]

Eine Einheit setzt in 3 min bei pH 6,9 und 20 °C 1,0 mg Maltose aus Stärke frei.

The protein content and enzymatic activity are determined on a lot to lot basis. Each value is reported in the specific Certificate of Analysis. The protein content range is 5 - 15% and the enzymatic activity range is 300 - 1500 units per milligram protein. A review of recent lots indicates typical protein concentrations of 7 - 13% and activities of 1350 - 1450 units per milligram protein. As mentioned in the 'DESCRIPTION' section, this product is a lyophilized powder containing (NH4)2SO4, sodium citrate and sodium chloride. This product is not tested for purity. However, buffer salts are expected to be the bulk of the non-protein content.
