Microtubules are involved in a wide variety of cellular activities ranging from mitosis and transport events to cell movement and the maintenance of cell shape. Tubulin itself is a globular protein which consists of two polypeptides (alpha and beta tubulin). Alpha and beta tubulin dimers are assembled to 13 protofilaments that form a microtubule of 22 nm diameter. Tyrosine ligase ads a C-terminal tyrosin to monomeric alpha tubulin. Assembled microtubules can again be detyrosinated by a cytoskeleton associated carboxypeptidase. Detyrosinated alpha tubulin is referred to as Glu-tubulin. Another post-translational modification of detyrosinated alpha tubulin is C-terminal polyglutamylation which is characteristic for microtubules in neuronal cells and the mitotic spindle.

Expected to cross-react with mammals, sea urchin, plants, not yeast (Paturle-Lafanechere, L., 1991).
The antibody specifically recognizes the detyrosinated form (Glu tubulin) of the tubulin alpha chain (Bianchi, M., 2006; Goulimari, P., 2005).

Peptide corresponding to the C-terminal of Glu tubulin.

Anti-Tubulin Antibody, Detyrosinated is an antibody against Tubulin for use in IC, IH(P) and WB.
Immunohistochemistry:
A previous lot was used on either frozen or paraffin embedded, fixed tissue (Gundersen, G.G., 1984): 1:200-1:500 dilution.

Optimal working dilutions must be determined by the end user.

Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
