General description

Amyloid-β-Protein wird aus dem Amyloid-β-Präkursor-Protein (APP) gebildet. Es besteht aus zwei C-terminalen Varianten, z. B. Aβ 1-42 mit langer Kette und Aβ 1-40 mit kurzer Kette. APP ist auf dem menschlichen Chromosom 21q21.3 lokalisiert.

Application

Amyloid-β-Proteinfragment 1-42 wird wie folgt verwendet:
  • zur Herstellung von Aβ-1-42-Oligomer[1]
  • in Western-Blot-Analyse
  • zum Störungstest von immunomagnetischem Reduktions(IMR)-Plasma-Aβ42-Assay
  • zur Untersuchung der Wirkung von Resveratrol auf Aβ-1-42-induzierte Beeinträchtigung von räumlichem Lernen, Erinnerung und synaptischer Plastizität
  • zur Untersuchung der Wirkung von Aβ in Epithelzellkulturen

Biochem/physiol Actions

Das am häufigsten vorkommende Fragment von Amyloid-β-Protein im Gehirn von Patienten mit Alzheimer′-Krankheit und Down′-Syndrom.
Amyloid-β-Proteinfragment 1-42 (Aβ 1-42) weist antioxidative und neuroprotektive Eigenschaften auf. Eine Ansammlung von Amyloid-β-Protein wird mit Alzheimer′-Krankheit (AD) und Down-Syndrom in Verbindung gebracht. Aβ 1-42 reguliert den Cholesterintransport und kann als Transkriptionsfaktor agieren. Es kann entzündungshemmende und antimikrobielle Eigenschaften aufweisen.

    1. This product can be reconstituted in ammonium hydroxide, pH >9, to a minimum of 1 mg/mL, or in DMSO at 0.4–4 mM.   2. After dissolving in DMSO, I want to dilute it with saline (finally 1ml/1mg) and use it for i.c.v. injection in mice. How many µl of 0.4-4 mM DMSO per 0.1 mg should I dissolve in?

    1. This product is soluble in DMSO at 0.4 - 4 mM, as stated in the product datasheet. This equates to 1.8 - 18 mg/mL in DMSO.   3. What is the difference between the AB peptide (1-42)(PP69) and AB protein fragment (1-42)(A9810)?

    1. These products are functionally the same, but are different brands. The PP69 is a Calbiochem product, while the A9810 is from Sigma-Aldrich. The testing specifications are slightly different, however both offer a minimum purity of 95%. The A9810 is lyophilized from a solution of water and TFA. The PP69 is lyophilized from a solution of 50 mM Tris-HCl. Please see the links below to review the product datasheet for each:

  4. It is instructed to dissolve the product in Ammonium Hydroxide solution, but what should be the molarity / Concentration of the ammonium hydroxide solution

    1. This product can be solubilized in ammonium hydroxide at a pH greater than 9. The concentration is not reported. It is recommended to prepare at a minimum 1 mg/mL concentration.

  5. How much ammonium hydroxide (pH >9) should be added to the 0,1 mg vial ? Is it possible to vortex the vial ?

    1. It is recommended to prepare a solution at a minimum of 1 mg/mL of ammonium hydroxide. Vertexing the vial is not necessary.

    1. As per the product information sheet, this product is soluble in ammonium hydroxide, pH >9, or in DMSO at 0.4–4 mM. Solutions can then be diluted into aqueous buffers such as10 mM sodium phosphate for use. Other reported solvents include hexafluoroisopropanol, acetonitrile, and acetic acid.
