Conn's Biological Stains, 10th Edition, page 250 states, "entry of the dye into cells may be achieved by direct insertion of the hydrophilic calcein, or by the uptake of calcein AM with intracellular formation of calcein."

Calcein is a calcium-dependent fluorescent molecule. It is mainly required to study bone metabolism under in vivo conditions and helps in the staining of pit area under in vitro conditions. Calcein is used for the fluorometric determination of calcium and EDTA (ethylenediaminetetraacetic acid) titration of calcium.
