(E/Z)-BCI hydrochloride has been used:

• to evaluate the role of the Duox-H₂O₂-Dusp6 derepression signaling pathway in heart regeneration[1]
• as an MKP-1 inhibitor to investigate the role of MKP-1 on bexarotent-induced effects
• as a dual specificity phosphatase 6 (DUSP6) inhibitor to study its effects on the proliferation of SKOV3 cells[2]

Biochem/physiol Actions

(E/Z)-BCI塩酸塩は、Dusp6(二重特異性ホスファターゼ6)とDusp1の阻害剤であり、FGFシグナリングを過剰活性化します。

BCIはDusp6のアロステリック阻害剤であり、ERK2基質の結合で生じるホスファターゼの触媒活性に対する刺激を、ホスファターゼドメインに作用することによって阻止します。特異性に関する研究から、BCIはDusp1を阻害する一方でDusp5は阻害せず、Cdc25B(細胞分裂サイクル25B)、PTP1B(プロテインチロシンホスファターゼ1B)やDusp3/VHRの活性をブロックすることが明らかとなっており、これらの事実からBCIの特異性は一連のMAPKホスファターゼに限られたものであることが示唆されています。

また、Dusp6がFGFシグナリングのフィードバック調節因子として機能することから、BCIはFGFシグナリングの過剰活性化を引き起こします。さらに、ゼブラフィッシュの体節形成期間にBCIがDusp6活性を阻害すると内皮系譜の心臓前駆体に拡大がみられるという報告があり、Dusp6は心蔵領域でFGFシグナリングのアテニュエーターとして、心臓組織の大きさを制御する役割があると言われています。
