Applicazioni

Il bromuro di etidio (EtBr) è il più usato tra i coloranti degli acidi nucleici per la PAGE o l′elettroforesi su gel di agarosio. La fluorescenza di EtBr aumenta di 21 volte quando si lega all′RNA a doppio filamento e di 25 volte quando si lega al DNA a doppio filamento così che con una bassa concentrazione del colorante (10 μg/mL) non è necessario decolorare il fondo. Il bromuro di etidio è stato usato in numerosi saggi fluorimetrici per gli acidi nucleici. È stato dimostrato il legame con il DNA a singolo filamento (anche se non altrettanto forte) e con il DNA a triplo filamento. Per la sua capacità di legarsi al DNA, il bromuro di etidio è un inibitore della DNA polimerasi.
Ethidium bromide solution has been used to deplete mtDNA to investigate mitochondrial damage signals in autophagy.[1] It has also been used to detect DNA damage using comet assay.[2]

Azioni biochim/fisiol

Il bromuro di etidio si frappone al DNA e all′RNA a doppio filamento e agisce come mutageno frameshift. Può essere usato anche insieme ad arancio di acridina per differenziare le cellule vitali, apoptotiche e necrotiche.

Ricostituzione

Per colorare un gel dopo l′elettroforesi, diluire un campione della soluzione madre con acqua a 0,5 μg/mL e incubare il gel per 15-30 minuti. La decolorazione, di solito non necessaria, si può eseguire in acqua per 15 minuti, se si deve ridurre il fondo. Si possono quindi rilevare le bande di DNA con l′illuminatore UV (lunghezza d′onda 254 nm). È anche possibile incorporare nel gel il bromuro di etidio, eseguire il tampone a 0,5 μg/mL e visualizzarlo immediatamente dopo l′elettroforesi.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.
