Application
Elastase aus Schweinepankreas wird zur proteolytischen Spaltung von UMP-Synthase (Uridin-5′-monophosphatsynthase) eingesetzt.[1] Es wird auch gemeinsam mit Kollagenase verwendet, um Gewebe in Einzelzellen aus thorakalem Medium, Bauchaortenmedium und Intima von Kaninchen zu dispergieren.[2]
Elastase aus Schweinepankreas wurde in einer Studie zur Identifizierung eines neuartigen Knochen-/Calciummetabolismus-Regulativs in Schweinepankreas eingesetzt. Elastase aus Schweinepankreas wurde außerdem in einer Studie zur Untersuchung der Wirkung auf einen spezifischen synthetischen Inhibitor der neutrophilen Elastase (ONO-5046) im Verlauf der akuten hämorrhagischen Pankreatitis bei Hunden eingesetzt.
Biochem/physiol Actions
Elastase hydrolysiert Elastin, das spezifische Protein elastischer Fasern, und verdaut Hämoglobin, Casein und Fibrin.
Elastase ist eine einzelne Peptidkette von 240 Aminosäureresten, die vier Disulfidbrücken enthält. Die Molekülmasse beträgt etwa 25,9 kDa. Das Enzym wird als ein inaktives Zymogen, Proelastase, das durch eine begrenzte Proteolyse durch Trypsin am N-Terminus zu seiner aktiven Form umgewandelt wird, synthetisiert.[3] Sie ist eine Serinprotease mit breiter Spezifizität. Sie spaltet Protein auf der Carboxylseite von kleinen hydrophoben Aminosäuren wie Ile, Gly, Ala, Ser, Val und Leu. Das Enzym hydrolysiert auch Amide und Ester wie N-Benzoyl-L-alaninmethylester.[3][4] Der optimale pH-Wert liegt bei 8,0–8,5. Es erfordert keinen Aktivator, wird aber durch Diisopropylfluorphosphat, Phenylmethylsulfonylfluorid, α2-Macroglobulin, α1-Antitrypsin, Sulfonylfluoride, p-Dinitrophenyldiethylphosphat und hohe Salzkonzentrationen gehemmt. Elastase kommt in Gewebe- und Zelldissoziationsverfahren ausgiebig zum Einsatz. Sie ist wirksam bei der Isolierung von Lungenzellen vom Typ II.
