Allgemeine Beschreibung

Adenosin-5′-triphosphat-dinatriumsalzlösung eignet sich zur Verwendung in molekularbiologischen Anwendungen wie Kinasereaktionen oder DNA-abhängige RNA-Polymerase-Transkription.

Anwendung

Adenosin-5′-triphosphat-dinatriumsalzlösung wird wie folgt verwendet:
  • Als Standard zum Beurteilen der extrazellulären ATP-Freisetzung aus Randzonenzellen der Stria vascularis[1]
  • Als Komponente der internen Pipettenlösung zum Etablieren einer Aufzeichnungselektrode zur Untersuchung der Neuritenstreckung[2]
  • im In-vitro-VPS34 (katalytische Untereinheit des Phosphatidylinositol-3-Kinase (Ptdlns3K)-Komplexes)-Aktivitätsassay[3]
  • als Komponente von Lyse- und Neuversiegelungspuffern für ATP-Studien[4]
  • bei der spektrofotometrischen Messung des H+-Transports in mit Fluoreszeinisothiocyanat (FITC)-Dextran beladenen A549- (menschliche alveoläre Adenokarzinom-Basalepithelzellen) und HeLa-Zellen[5]
  • zur Einleitung der Pryoptose in mit Lipopolysaccharid behandelten fibroblastenähnlichen Synoviozyten (FLS)[6]

Biochem./physiol. Wirkung

Adenosin-5′-triphosphat (ATP) ist ein wichtiger Energieträger in allen lebenden Organismen. Es besitzt ein hohes Phosphattransferpotenzial. ATP ist an mehreren biologischen Vorgängen wie Membrantransport, Muskelkontraktion und -synthese sowie dem Abbau von biologischen Molekülen beteiligt. Es spielt als intra- und extrazelluläres Signalmolekül eine Rolle in bestimmten zellulären Vorgängen wie der Zellmotilität, der Organentwicklung, der Neurotransmission und der Insulinsekretion.[7]

  1. How does the storage temperature relate to shipping conditions?

    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

    1. This preparation of ATP is not recommended for use as an ATP standard. The recommended ATP products for use as a standard in bioluminescence experiments is Product Code FL-AAS (preweighed vials of 1 mg each = 2 micromole).

  3. What is the binding affinity of ATP (Product No. A6559) for Mg++ and other metal ions?

    1. ATP has intrinsic metal binding affinity. The binding constant for various metals are (given as per mole): Mg++, 9554; Na+, 13; Ca++, 3722; K+, 8; Sr++, 1381; Li+, 25.  The ability of ATP to chelate magnesium results in effective Mg++ ion concentrations lower than the total Mg ion concentration.  For example, in a reaction with 1.5mM Mg++ and 0.2 mM ATP, the effective Mg ion concentration is ~1.3mM.

    1. This is a Molecular Biology grade ATP. It is certified free of DNase and RNase. This product is recommended for any typical molecular biology application.

    1. The purity of this product should be sufficient for use in T7 RNA polymerase-catalyzed in vitro transcription assay, but we do not test the product for use in that application in our quality control process.

    1. Aqueous solutions of ATP are stable for months when frozen in solution at -15 °C and for approximately one week at 0 °C.  As with most products, we guarantee the quality of this material for 1 year when stored unopened at the recommended storage temperature (-20°C in this case).

    1. 25UMO means 25 micromoles.  The 25 UMO size is equivalent to 250 μl of a 100 mM solution. This product is a 100 mM solution in water; the solution pH has been adjusted with Trizma base but contains no additional buffer or salts.
