Leucin-Aminopeptidase gehört zur Familie der Metallproteasen.

Application

Leucin-Aminopeptidase, mikrosomal aus der Schweine-Niere wird bei der Isolierung von Neprilysin (NEP) aus Nagetierhirn-C5 und für den Peptidaufschluss eingesetzt.

Biochem/physiol Actions

Leucin-Aminopeptidase besitzt eine breite Substratspezifizität. Sie hydrolysiert Peptide bis zum Prolinrest und baut nach Prolin nicht weiter ab.

Physical form

Suspension in 3.5 M (NH₄)₂SO₄-Lösung, pH 7.7, enthält 10 mM MgCl₂

Other Notes

Eine Einheit hydrolysiert 1.0 μmol L-Leucin-p-nitroanilid pro Min. zu L-Leucin und p-Nitroanilin bei pH 7.2 und 37 °C. (Bei 25°C, wird ca. 40% der bei 37 °C gefundenen Aktivität erreicht.) Die Aktivität bei Verwendung von L-Leucin-p-nitroanilid als Substrat ist 2-5 Mal höher als bei der Verwendung von L-Leucinamid als Substrat.
