Application
Das Enzym von Sigma wurde für die Entwicklung eines homogenen zeitaufgelösten Fluorszenz-Assays (HTRF) zur Messung der Carboxypeptidase-B-Aktivität in einem miniaturisierten Screening-Format mit hohem Durchsatz verwendet. Es wurde auch für die Beurteilung der Auswirkung von C-terminalen Lysinen in humanem, an Bifidobacterium bindendem Plasminogen verwendet. Die Wirkung der Behandlung mit Carboxypeptidase B, welche eine C-terminale lysinspezifische Endopeptidase ist, wird mittels flusszytometrischer Analyse gemessen.
Carboxypeptidase B wurde in einer Studie zur Entwicklung eines nicht-invasiven Schwangerschaftstests zur Verwendung bei in Gefangenschaft gehaltenen und wildlebenden Eisbären verwendet. Carboxypeptidase B wurde in einer Studie zur Identifizierung neuer potenzieller Biomarker für akute Pankreatitis verwendet.
Biochem/physiol Actions
Carboxypeptidase B ist ein proteolytisches Enzym, das zu einer raschen Hydrolyse von Peptidbindungen zur Freisetzung bestimmter Carboxyl-terminaler basischer Aminosäuren aus Peptiden und Proteinen fähig ist. Seine Molekularmasse beträgt 34,300 ±600 Da. Es enthält ein nicht dialysierbares Gramm Zinkatom pro Mol. Die Enzymaktivität wird gehemmt durch die Metallchelatbildner 1,10-Phenanthrolin, 8-Hydroxyquinolin-5-sulfonsäure und 2,2′-Dipyridyl.
