Questions

  1. What is the extinction coefficient for this 80 mg/mL protein standard?

    1 answer
    1. This product is a mixture of human serum albumin (HSA) and human gamma-globulin, the latter of which is essentially all human IgG. The E1% (at 280 nm) values for HSA and human IgG are 14 and 5.3, respectively. The lot-specific extinction coefficient for the mixture can be calculated using the protein concentrations provided the certificate of analysis (CofA)._x000D_

      https://www.sigmaaldrich.com/certificates/sapfs/PROD/sap/certificate_pdfs/COFA/Q14/P8119-BULKSLCN8399.pdf_x000D_
      The CofA reports an HSA concentration of 4.9 g/dL, and (by difference) a gamma-globulin concentration of 3.3 g/dL. The extinction coefficient at 280 nm for this 8.2 g/dL (82 mg/mL) lot is 86. Here is the calculation: (4.9 X 14) + (3.3 X 5.3) = 86.09, which would round to 86. If the standard were diluted by a factor of 8.2 to give a 1 g/dL or 1% solution, the E1% would be 10.5.
