FUNCTION: SwissProt: P29972 # Forms a water-specific channel that provides the plasma membranes of red cells and kidney proximal tubules with high permeability to water, thereby permitting water to move in the direction of an osmotic gradient.
SIZE: 269 amino acids; 28526 Da
SUBUNIT: Homotetramer.
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Expressed in a number of tissues including erythrocytes, renal tubules, retinal pigment epithelium, heart, lung, skeletal muscle, kidney and pancreas. Weakly expressed in brain, placenta and liver.
DOMAIN: SwissProt: P29972 Aquaporins contain two tandem repeats each containing three membrane-spanning domains and a pore-forming loop with the signature motif Asn-Pro-Ala (NPA).
SIMILARITY: Belongs to the MIP/aquaporin (TC 1.A.8) family.
MISCELLANEOUS: Pharmacologically inhibited by submillimolar concentrations of mercury.

Highly purified peptide corresponding to residues 242-260 of human Aquaporin 1 (SwissProt accession number P29972).

Recognizes Aquaporin 1.

SPECIES REACTIVITIES: The epitope specific for Aquaporin 1 is identical in human, rodent, bovine and only slightly different in frog (18/19 aa). Reactivity with other species has not yet been tested.

Zastosowanie

Western blotting: 1:500-1:1000 on rat kidney membranes.

Immunohistochemistry

Dilutions should be made using a carrier protein such as BSA (1-3%).

Optimal working dilutions must be determined by the end user.

Postać fizyczna

Purified immunoglobulin. Lyophilized from PBS, pH 7.4, containing 0.025% sodium azide. Reconstitute with 200 μL of sterile distilled water. Centrifuge antibody preparation before use (10,000 x g for 5 min).

Przechowywanie i stabilność

Maintain lyophilized material at -20°C for up to 6 months after date of receipt. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Inne uwagi

Included free of charge with the antibody is 40 μg of control antigen (lyophilized powder). Reconstitute with 100 μL of deionized water. For negative control, preincubate 1.0 μg of purified peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.

The epitope specific for Aquaporin 1 is identical in human, rodent, bovine and sheep and only slightly different in canine (18/19 amino acids) and frog (14/19 amino acids).
