應用

推荐使用人成纤维细胞的层粘连蛋白作为1-2 μg/cm2细胞培养基基质。 最佳的浓度的确取决于细胞类型以及应用或研究目标。

生化/生理作用

层粘连蛋白是动物组织中结构性支架的组成部分。它们通过内功素和基底膜聚糖与IV型胶原结合,并通过整合素受体、dystroglycan糖蛋白复合物以及Lutheran血型糖蛋白与细胞膜结合。层粘连蛋白具有胶原结合、细胞粘附、肝素结合和神经突向外生长片段的活性结构域。层粘连蛋白支持多种细胞类型的生长和分化,包括表皮、内皮、神经、肌肉和肝脏细胞。

成分

层粘连蛋白是细胞外基质多结构域三聚体糖蛋白,并且是支持粘附、增殖和分化的基底层的主要非胶原组分。 层粘连蛋白由A、B1和B2 链组成,它们通过许多二硫键连接。该层粘连蛋白产物由人成纤维细胞和上皮细胞在共培养系统中产生,然后通过生物化学方法纯化。

注意

建议将此产品储存在-70°C,它可以其中稳定保存两年。

準備報告

该产品在TBS中以 0.5mg/mL的浓度供应。在使用前在2-8°C下将该溶液缓慢解冻以避免形成凝胶。如果在解冻之前的任何时候过早加热层粘连蛋白,则可能形成凝胶。此外,即使在 -20C下储存也可能导致凝胶形成或活性显著下降,或出现这两种现象。用作包膜时,在HBSS中稀释,以最小体积涂覆培养物表面并在37°C 下孵育 1-2 小时。 在接种细胞前使用HBBS 洗涤 3 次。层粘连蛋白包膜可在2-8°C储存一个月。
