纤连蛋白(FN)是糖蛋白,5%-9%的糖类以 N-和 O-连接寡糖的形式存在。 属于配体糖蛋白家族。FN基因位于人类染色体2q35。FN以两种主要的同工型存在,即,基于血浆的可溶形式和基于细胞外基质(ECM)的不溶形式。纤连蛋白结构包含FNI、FNII和FNIII重复序列的重复单元。它还具有血纤蛋白结合位点,以二聚体形式存在。FN1基因的可变剪接导致产生许多同工型。该纤连蛋白是用全人类生产系统在人细胞中生产,不含血清。该人类细胞表达系统可实现与人类蛋白相仿的糖基化和折叠,通常可为培养物中的蛋白提供更好的稳定性支持。

應用

上皮细胞,间充质细胞,神经元细胞,成纤维细胞,神经嵴细胞,内皮细胞。
推荐用作细胞培养基质,1-5μg/cm 2 或 0.5-50μg/ml。最佳浓度取决于细胞类型以及应用或研究目的。
人纤连蛋白用于包被细胞培养板,培养内皮祖细胞。它还作为Dulbecco′s改良Eagle′培养基(DMEM)成分,用于胶质母细胞瘤细胞的 体外 人工基膜侵袭试验。

生化/生理作用

纤连蛋白(FN)对胚胎发育至关重要,介导细胞的粘附和迁移功能。介导鼻咽癌(NPC)细胞凋亡。血浆FN调节血栓形成。纤连蛋白可作为整联蛋白受体家族 的配体,包含胶原蛋白、糖蛋白和糖胺聚糖结合位点。 FNI基因突变造成纤连蛋白的可溶形式和不可溶形式失衡,导致遗传性肾脏疾病-肾小球病中的肾小球沉积。FN1基因的错义突变与骨骼发育不良疾病-脊柱干骺端发育不良有关。
