ラミブジンは、強力なヌクレオシドアナログ逆転写酵素インヒビター(nRTI)です。シチジンのアナログで、HIV逆転写酵素の両タイプ(1と2)ならびにB型肝炎逆転写酵素を阻害することができます。活性型になる前に、三リン酸塩にリン酸化される必要があります。3TC三リン酸塩も、細胞DNAポリメラーゼを阻害します。
