ヒトPC4は非TAF転写共役因子であり、in vitroで、ほとんどの試験済み活性化因子を介してRNAポリメラーゼIIによる活性化因子依存性転写を開始させます。PC4は、転写活性化因子と基本転写装置の相互作用を介して機能します。PC4は、in vitroとin vivoの両者でカゼインキナーゼIIリン酸化により負に調節されています。PC4は一本鎖DNAに強力に結合し、この一本鎖DNA結合活性に欠かせない領域はアミノ酸残基77位付近にマッピングされました。第77位における1個のアミノ酸置換(FからP)により、二本鎖DNAと一本鎖DNAへの結合活性は、いずれも阻害されます。
