クリプトタンシノンは、アジア地域の薬草であるタンジン(Salvia Miltiorrhiza Bunge)の根から単離されたキノイド型ジテルペンです。この化合物は、近年、STAT3の強力なインヒビターであることが発見されました。クリプトタンシノンは、JAK2に非依存的なメカニズムを通じてSTAT3のTyr705リン酸化を急速に阻害します。また、単量体STAT3と結合することでSTAT3の二量体化と転写調節活性をブロックし、STAT3活性化細胞株を選択的に阻害します。この化合物に関して、シクロオキシゲナーゼIIの活性やエンドセリン-1の発現を阻害して炎症を抑制するということは、以前より報告されていました。東洋医学において、タンジンの乾燥根は伝統的に循環障害、肝疾患、冠動脈心疾患、肝炎、慢性腎不全の治療に広く利用されています。
クリプトタンシノンは、特異的なSTAT3阻害剤です。TAT3の二量体化を防止します。
