ジチオトレイトール（DTT）は、タンパク質およびアミノ酸システインに見られるジスルフィド（SH）還元剤です。ジスルフィド結合の還元は、6つのメンブレンリングを形成してチオール-ジスルフィド交換を開始することによります。βMEの約7倍の強度を持つ還元剤であるため、低濃度で効果を発揮します。

アプリケーション

DTTは以下に使用されています：
  • ウシミルクの主なアレルゲンである、αs1-カゼインの還元およびアルキル化における反応物質の1つとして。[1]
  • 精子の脱膜化および再活性化のための培地の成分として。[2]
  • バイオ触媒の活性部位を効果的に保護するチオールとしての酵素の安定化を維持するため。[3]
  • スルフヒドリル基を有するN-エチルマレイミドの反応特異性を調べるための還元剤として。[4]
  • プロテオミクス分析における、質量分析法向け溶液中タンパク質分解物として[5]
  • タンパク質定量のためのバッファー成分として、洗浄バッファー、溶解バッファー、サンプルバッファー、およびタンパク質溶出バッファーを調製するため[6][4]
SH基を還元状態に保つ試薬です。ジスルフィドを定量的 に還元します。SDS-PAGEのサンプルバッファーに加えることで、タンパク質のジスルフィド結合の還元に効果を発揮します。また、架橋剤であるN,N′-ビス(アクリロイル)シスタミンのジスルフィド結合を還元してポリアクリルアミドゲルのマトリックスを切り離す目的でも用いられます。2-メルカプトエタノールと比較して、刺激性および毒性が抑えられています。通常、2-メルカプトエタノールの使用濃度(5% v/v、700 mM)の7分の1濃度(100 mM)で効果を発揮します。

生物化学的／生理学的作用

ジチオトレイトール（DTT）は、化学的ペプチド合成およびチオールタンパク質の生化学的調製に幅広く応用されています。また、タンパク質フォールディングおよび酵素活性などのタンパク質化学の研究にも使用されています。DNA修飾を止めるための酵素安定剤としても機能します。DTTは、チオール-ジスルフィド交換反応を特異的に媒介することにより、生体分子の分子内または分子間ジスルフィド結合を完全に還元します。この反応は、チオールの形成とDTTの環状ジスルフィドをもたらします。[7]

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   3. How long can the powder be left out a room temperature?

    1. The long term storage recommendation for this product is refrigerated at 2-8°C, but is shipped at ambient temperature. Refrigerated products that ship at ambient temperature are stable for up to 2 weeks to allow for transit time. Items left at room temperature for short periods will not be adversely effected.

  4. What is the shelf life of this product once the stock solution is made up please?

    1. A solution of DTT in Hepes buffer, pH 7.75 is stable for one week at 2 to 8 °C if the container is tightly sealed and the solution is protected from atmospheric oxygen by argon or nitrogen.
