アルファ-シヌクレイン（UniProt P37840、別名NACP、ADアミロイドの非Aベータ成分、アミロイド前駆体の非A4成分、シヌクレインアルファ-140）は、ヒトSNCA（別名NACP、PARK1、PARK4）遺伝子（Gene ID 6622）によってコードされています。異常な凝集体は、アルツハイマー病（AD）と前頭側頭葉変性症におけるタウ神経原線維変化（NFT）、およびパーキンソン病（PD）とレビー小体型認知症（DLB）におけるα-シヌクレイン（α-syn）レビー小体（LB）などの多数の神経変性疾患に共通する特徴です。アルファ-シヌクレインは、シナプス前終末に濃縮されているリン脂質結合タンパク質であり、SNARE複合体の形成を促進し、シナプス機能を調節します。アルファ-シヌクレインは、PD、DLB、および多系統萎縮症（MSA）の特徴である病理学的封入体の主成分です。カゼインキナーゼ-1（CK-1）とCK-2はともに、アルファ-シヌクレインのSer129でのリン酸化を触媒できます。アルファ-シヌクレイン封入体には、Ser129でリン酸化されたアルファ-シヌクレインが存在しています。
特異性
クローン81Aは、CK1-およびCK2-触媒アルファ-シヌクレインSer129リン酸化を検出しましたが、CK1-触媒-アルファ-シヌクレインSer87リン酸化と 非リン酸化-シヌクレインは検出しませんでした（Waxman, E.A., et al. (2008). J. Neuropathol. Exp. Neurol. 67(5):402-416）。
