Applicazioni

Lectin from Phaseolus vulgaris (red kidney bean) has been used:
  • for the stimulation of cluster of differentiation (CD4+) T cells[1]
  • for the activation of primary peripheral blood lymphocytes[2]
  • for the stimulation of peripheral blood mononuclear cells[3][4]

Azioni biochim/fisiol

PHA consists of two molecular species, an erythroagglutinin (PHA-E) which has low mitogenic activity and high erythroagglutinin activity, and leucoagglutinin (PHA-L) which has high mitogenic and leucoagglutinating activity, but very low erythroagglutinating activity.
PHA-E is not blood group specific, but agglutination can be inhibited by certain oligosaccharides. PHA-P is the protein form of PHA prior to separation and purification of erythroagglutinin and leucoagglutinin. PHA-M is the mucoprotein form. Conjugates are prepared from the corresponding purified lectins.

Questions

1–7 of 7 Questions
  1. What do the designations -P, -L, -M, and -E represent in the names of PHA products?

    1 answer
    1. Generally, PHA-P or PHA-L is utilized to stimulate lymphocytes, although PHA-M will also work, but not to the same degree. The most active form is PHA-L (the mitogenic concentration is < 5μg/ml). PHA, or Phytohemagglutinin, comprises two molecular species:

      1. Erythroagglutinin (PHA-E), which demonstrates low mitogenic activity and high erythroagglutinating activity.

      2. Leucoagglutinin (PHA-L), which possesses high mitogenic and leucoagglutinating activity, accompanied by very low erythroagglutinating activity.

PHA-P represents the protein form of PHA before the erythroagglutinin and leucoagglutinin components are separated and purified. PHA-M refers to the mucoprotein form of Phytohemagglutinin.

      Helpful?

  2. How should the PHA solution using Phytohemagglutinin PHA-P powder be prepared for cell culture?

    1 answer
    1. The stock solution of this product can be prepared in phosphate buffered saline at pH 6.8, resulting in a clear to slightly hazy solution at a concentration of 5 mg per mL. This stock solution can then be diluted in cell culture medium to a working concentration of 10 micrograms of lectin per mL for use in peripheral blood cell culture.

      Helpful?

  3. What is the shelf life once reconstituted in PBS?

    1 answer
    1. After reconstitution in PBS, this product can be stored at 2-8°C for 2-4 weeks. PHA loses approximately 30% activity after 1 month. Stock solutions of at least 1 mg/mL can be stored in frozen aliquots. Do not dilute to the working concentration until ready to use. Vortex stock solutions prior to use.

      Helpful?

  4. What is the mitogenic activity of Product L1668, Lectin from Phaseolus vulgaris (red kidney bean) - Phytohemagglutinin PHA-P, lyophilized powder?

    1 answer
    1. The minimum specification for mitogenic activity of this product is  < or = 10 micrograms lectin/mL of medium. The reported specification is PASS. A specific value is not given.

      Helpful?

  5. What is the Department of Transportation shipping information for this product?

    1 answer
    1. Transportation information can be found in Section 14 of the product's (M)SDS.To access the shipping information for this material, use the link on the product detail page for the product.

      Helpful?

  6. How can Product L1668, Lectin from Phaseolus vulgaris (red kidney bean) - Phytohemagglutinin PHA-P, lyophilized powder, be dissolved?

    1 answer
    1. Product No. L1668 is solubility tested using 5 mg per mL of phosphate buffered saline (PBS), pH 6.8; this yields a clear to slightly hazy, colorless to light yellow solution.

      Helpful?

  7. What is the solution stability for Product L1668, Lectin from Phaseolus vulgaris (red kidney bean) - Phytohemagglutinin PHA-P, lyophilized powder?

    1 answer
    1. The solution stability was not evaluated by Sigma-Aldrich; however, references indicate media containing dilute PHA can be stored refrigerated for 2-4 weeks. PHA loses approximately 30% activity after 1 month. Stock solutions can be stored in frozen aliquots.
