Die Verwendung von SIGMAFAST Proteaseinhibitor-Tabletten mit HIS-Select® Produkten oder anderen Produkten für die immobilisierte Metall-Affinitätschromatographie (IMAC) wird nicht empfohlen. Diese Tabletten enthalten den Metallchelator EDTA, der möglicherweise Metallionen aus diesen Affinitätsprodukten entfernen kann.
SIGMAFAST™ Proteaseinhibitor-Tabletten sind eine wasserlösliche Tablettenformulierung, die einen Breitspektrum-Cocktail von Proteaseinhibitoren enthält, darunter AEBSF, E-64, Bestatin, Leupeptin, Aprotinin und EDTA. Eine Tablette liefert 100 ml Cocktail und kann Proteasen in bis zu 20 g Zellextrakt hemmen.
Spezifität
Hemmt Serin-, Cystein-, Asparagin- und Metallproteasen.
Anwendung
Diese Tabletten wurden für den allgemeinen Gebrauch optimiert und geprüft. Sie wurden in verschiedenen Anwendungen eingesetzt, einschließlich Sammeln von Tracheen in Kombination mit eiskaltem Radioimmunpräzipitationsassaypuffer zum Bestimmen des Schicksals von trachealen Basalzellen und Homogenisieren von Rattenlungengewebe für die Western-Blot-Analyse.
SIGMAFAST Proteaseinhibitor-Tabletten wurden wie folgt verwendet:
- zum Waschen von unbearbeitetem Sperma für die Membranproteinanalyse[1]
- in einer Lösung zur Inkubation von CD4+ T-Zellen, um Gesamtzelllysate für Western Blot-Analysen zu gewinnen[2]
- als Komponente von NP-40-Puffer zum Homogenisieren des Rückenmarksgewebes im Bereich des Epizentrums einer Verletzung zur Durchführung einer Western-Blot-Analyse[3]
- als Komponente im Puffer für Radioimmunpräzipitationsassays (RIPA) zum Homogenisieren des Rückenmarksgewebes im Bereich des Zentrums einer Verletzung, um Neuregulin-1-Beta1 (Nrg-1ß1) in Proben des Rückenmarkgewebes mittels Enzyme-linked Immunosorbent Assay (ELISA) nachzuweisen[3]

Eine Tablette ergibt 100 ml Cocktail. Ein ml wird zur Hemmung von Proteasen, die 1 mg USP-Pankreatin entsprechen, empfohlen. Eine Tablette wird zur Hemmung der Proteasen in maximal 20 g Zellmasse empfohlen.
