General description

Cholera-Toxin gehört zur Toxinfamilie der AB5-Untereinheit. Es besteht aus einer einzelnen A-Untereinheit, die für die ADP-Ribosylierung verantwortlich ist, und fünf B-Untereinheiten, die einen pentameren Ring bilden und Funktionen bei der Bindung an Zelloberflächenrezeptoren und der Internalisierung aufweisen. Die A-Untereinheit katalysiert die ADP-Ribosylierung der α-Untereinheit des stimulierenden G-Proteins (Gαs), was zu einer erhöhten Adenylylcyclaseaktivität und einem erhöhten cAMP-Spiegel führt. Außerdem ADP-ribosyliert es Transducin und Tubulin.

Cholera-Toxin und seine A-Untereinheit werden aufgrund ihrer Auswirkungen auf die Adenylylcyclase häufig zur Untersuchung von Signalübertragungsmechanismen verwendet. Darüber hinaus wirkt Cholera-Toxin als Adjuvans, indem es die Reaktionen von B-Lymphozyten und Typ-2-T-Helferzellen durch Hemmung der Interleukin-12-Produktion stimuliert. Die nicht toxische B-Untereinheit (CTB) bindet an das Gangliosid GM1 und eignet sich daher für die Markierung von Mikrogliazellen in der neurologischen Forschung. Es hat sich auch als ausgezeichneter Tracer für axonale Transportstudien mit immunhistochemischen Methoden erwiesen.

Application

Das Cholera-Toxin von Vibrio cholerae wird in Toxin-Neutralisationsassays zur Beurteilung der Zytotoxizität von Zellen verwendet.
Choleratoxin aus Vibrio cholerae wird als positive Kontrolle im cAMP(cyclisches AMP)-Assay für Enterotoxine eingesetzt. Es wird als Ergänzung zu Zellkulturmedien von Primärtumoren und Epithelzellen eingesetzt.

Biochem/physiol Actions

Toxin, das aus einer A-Untereinheit (27 kDa) besteht, die von fünf B-Untereinheiten (jeweils etwa 12 kDa) umgeben ist, die das Toxin an Ganglosid GM1 auf der Zelloberfläche binden.

    1. Dilution of the cholera toxin to 10 ng/mL in water is possible. Storage is recommended at 2-8 °C to avoid freezing, as stability in solution at -20 °C has not been determined.

    1. As listed in the Product Data Sheet, the lyophilized powder must be stored at 2–8°C. The product, as supplied, is stable for 3 years when stored properly.

  5. Hey, Can this product be reconstituted to a solution and administered to mice for systemic upregulation of immune cells?

    1. This product has not been tested for use in animal models, however, the preparation does contain sodium azide as a preservative. Azide is reported to have an LD50 of 19 mg/kg in mouse by I.V. route. This is not a sterile preparation, solutions would require filter sterilization prior to in vitro or in vivo use.     1. It is not necessarily sterile, but the solution can be filter sterilized using a 0.2 micron pore size filter.
