Descrição geral

A hidroxiprolina (4-hidroxiprolina) é um aminoácido não proteinogênico[1] formado pela hidroxilação pós-tradução da prolina. A hidroxiprolina é um componente importante do colágeno, no qual tem a função de estabilizar a estrutura helicoidal.[2] Dado que, em sua maior parte, a hidroxiprolina está restrita ao colágeno, a medição dos níveis de hidroxiprolina pode ser usada como um indicador do teor de colágeno. As condições que aumentam a taxa de síntese e degradação do colágeno podem aumentar os níveis de hidroxiprolina no soro e na urina. Os níveis de hidroxiprolina no soro e na urina podem ser usados como marcadores de reabsorção óssea.[3][4]

Aplicação

O kit de ensaio de hidroxiprolina foi usado para determinar a concentração de hidroxiprolina e colágeno.[5][6][7]

Adequação

Adequado para a detecção de hidroxiprolina em sobrenadantes de cultura de tecidos e células, urina, plasma, soro e outras amostras biológicas.

Princípio

No kit de ensaio de hidroxiprolina, a concentração de hidroxiprolina é determinada pela reação da hidroxiprolina oxidada com 4-(dimetilamino)benzaldeído (DMAB), que gera um produto colorimétrico (560 nm) proporcional à hidroxiprolina presente.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  4. Can this kit be used to quantify 3D collagen scaffolds or for coatings on scaffolds? Has it been done?

    1. This type of sample has not been tested with this assay kit. It is suggested to consult the scientific literature to determine if the specific sample type has been reported for this type of usage.

  5. 1. Are heart samples that have been flash frozen suitable for use with MAK008-1KT?

    2. The manual mentions 10 mg of tissue, but does it need to be an exact measurement? Is it challenging to accurately weigh the frozen tissue?

    1. 1. Flash frozen tissue can be used with the kit. The recommended approach is to homogenize the tissue with room temperature water. Allow the tissue to thaw slightly by leaving it in the water for a few seconds before homogenization.

      2. The kit is optimized for use with 10 mg of tissue homogenate supernatant. Slight variations below or above 10 mg will not affect the assay significantly. However, it is advisable to avoid large differences in the weight or mass of the tissue.

  6. Is it possible to carry out the 90-minute incubation at 60 °C, as mentioned in the protocol after adding the DMAB Reagent, using a hot plate? and, should the plate be covered during this incubation step?

    1. Yes, it is possible to perform the incubation on a hot plate. It is recommended to cover the plate during this process.

  7. Protocol recommends 10 mg tissue or cells for sample preparation. Is there a minimum quantity of cells required for this kit?

    1. It is recommended to start with 1-2 million cells per sample. However, determining a specific minimum amount is challenging because the level of hydroxyproline can vary significantly between different sample types.
