Beschreibung

General description

Ein zellpermeable Pyrimidinverbindung, die die GDP/GTP-Austauschaktivität von Rac 1 spezifisch und reversibel hemmt, indem sie die Interaktion zwischen Rac1 und Rac-spezifischen Guanin-Nukleotid-Austauschfaktoren (GEFs) Trio und Tiam1 (IC₅₀ ~50 µM) behindert. Hemmt nachweislich die Rac-1-vermittelten Zellfunktionen von NIH3T3- und PC-3-Zellen (wirksame Dosis ~50 bis 100 μM). Weist keine Wirkung auf die Cdc42- oder RhoA-Aktivierung auf und betrifft nicht die Rac1-Interaktion mit BcrGAP oder PAK1. Reduziert die TRAP-induzierte und kollagenstimulierte Thrombozytenaggregation (IC₅₀ = 50 mM bzw. 64 mM).
