一般說明

胰岛素-转铁蛋白-亚硒酸钠(ITS)添加剂对于很多细胞类型来说都是最为重要的3种生长因子。胰岛素是所有原代细胞和细胞系的无血清培养基配方成分。在无血清培养基中用于促进细胞生长、增加脂肪酸和糖原合成。转铁蛋白是许多类型细胞必需的生长因子。硒常用于细胞生长优化。ITS可作为补偿,代替相当可观的细胞生长所需的血清用量。[1]ITS也可作为抗氧化剂,支持体内生产(IVD)胚胎的生长。其中,胰岛素可促进胚胎生长和代谢。转铁蛋白介导胚胎中的铁转运。亚硒酸钠保护细胞免于氧化损伤并抑制脂质过氧化。[2]ITS可用于人软骨细胞单层培养。[1]
胰岛素-转铁蛋白-亚硒酸钠补充剂以最佳比例包含无血清培养基中最重要的促生长成分。

應用

胰岛素-转铁蛋白-亚硒酸钠补充剂在细胞培养培养基中用作基础生长因子补充剂,向其中添加其他细胞类型特异性营养素和生长因子。[3][4][5]

準備報告

工作浓度:5 μg/mL胰岛素、5 μg/mL转铁蛋白、5 ng/mL亚硒酸钠(3.0 x 10-8 M)

储存条件(工作溶液):-15至-25℃。
制备适当的等分试样并避免反复冻融。

重構

胰岛素-转铁蛋白-亚硒酸钠补充剂应在5 ml或25 ml无菌双蒸水(1000 x 浓缩储备液)中复溶。
。用培养基进一步稀释。
