一般描述

牛胰腺脱氧核糖核酸酶I（DNase I）是DNA小沟相互作用的核酸酶，特异性相对较低。该蛋白质由两个中心β 折叠构成，而每个折叠由六个β-链所组成。该结构被大量的环以及α-螺旋区域包围。该酶与核酸外切酶III具有结构相似性。[1]DNase I是研究得最透彻的哺乳动物来源的内切酶之一。它是一种双链特异性内切核酸酶，需要二价阳离子激发最大活性。[2]

应用

蛋白质（如膜蛋白）的分离操作。
牛胰腺脱氧核糖核酸酶I（DNase I）已被用于：
  • 分离肺、皮肤和肿瘤样品中的细胞[3][4][5][6]
  • 从抗DNase I的活细菌细胞中提取细菌DNA †

单位定义

制备说明

激活剂：Ca2+ (0.12 mM)与Mg2+结合在一起可增强活性 [Laskowski (1971); Melgar及Goldthwait (1968)].
工作浓度：1 mg/ml
在溶解过程中请勿涡旋！
储备条件 (工作溶液)：当在水中重悬时，溶液可在2至8 °C下保存2至3天。在溶解过程中请勿涡旋。注：至少以1 mg/ml溶解。
