애플리케이션

Achromopeptidase from bacteria may be used:

• in the homogenization step for genomic DNA extraction from microbial biofilm[1]
• in the lysis of gram-positive strains([2]
• for DNA extraction from Nocardia strains[3]

Achromopeptidase is useful for lysis of Gram-positive bacteria that are resistant to lysozyme. Achromopeptidase, from Sigma, has been used for the isolation and molecular characterization of skin and soft-tissue bacterial infections[4]. Product A3422 is from bacteria and is a partially purified powder containing lactose.

생화학적/생리학적 작용

Achromopeptidase is a bacteriolytic enzyme and belongs to serine protease family.[5] It is effective for the lysis of gram positive bacteria compared to lysozyme.[6] Achromopeptidase activity is inhibited in the presence of heavy metals. Achromopeptidase based inhibition assay may be useful for screening the presence of heavy metals.[5]

Achromopeptidase is a lysyl endopeptidase that has a molecular weight of approximately 27 kDa. It works best between pH 8.5 -9. Approximately 500-1,500 μn/ml achromopetidase can be used to lyse cells at a density of OD₆₀₀ = 0.6 over 2 hours at 37 °C.
