アプリケーション

蛍光チオールアッセイキットは、サンプル中のチオールを示す脂質過酸化反応を求めるために使用されます。[1]

適合性

このキットは、血漿、尿、細胞抽出物などのさまざまな生体サンプル中のチオールの検出に適しています。このキットは、タンパク質中のチオール基の測定には最適化されていません。

原理

蛍光チオール定量キットは、低分子チオールの含有量を定量するための超高感度アッセイを提供します。独自のチオールセンサーは、チオール含有化合物と反応すると、強力な蛍光付加物（λex= 490 /λem= 520 nm）を生成します。さらに、チオール付加物の吸収スペクトルと発光スペクトルの両方がpHに依存しないため、このアッセイキットは非常に堅牢です。このキットは、100 μLのアッセイ容量中のわずか1ピコモルのシステインまたはGSH（10 nM）を検出できます。アッセイは利便性が高く、96または384マルチウェルプレート形式で実施できます。

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. Can this kit be used to quantify cysteines from proteins? If so, do I need to reduce disulfide bonds first? Also, does maleimide interfere with the assay?

    1. This kit detects reduced thiols such as reduced Glutathione (GSH) and Cysteine. In addition, the product may also be used to detect SH groups in the protein, provided that a standard with known SH groups is used, such as BSA (0.5-1 GH/molecule). The possible interference of Meleimide has not been investigated.

  4. Would MAK151 detect reduced thiols like GSH, Cysteine, and NAC, or does it detect total thiol? Additionally, does it detect total protein-SH or protein-S-S protein?

    1. MAK151 is capable of detecting reduced thiols like GSH, Cysteine, etc. It can also be used to detect SH groups in proteins, provided that known SH groups of the protein standard, such as BSA (0.5-1 GH/molecule), are available.
