ヘキソキナーゼは、ATP依存性リン酸化を触媒することによりアルドヘキソースおよびケトヘキソースをヘキソース-6-リン酸に変換し、多数の重要な生化学的経路の第一段階を触媒しています。哺乳動物組織では、グルコースはヘキソキナーゼの主要な基質です。[1]哺乳動物組織ではヘキソキナーゼには4つのアイソザイムがあり（HK-I、-II、-IIIおよび-IV）、動態および調節特性、さらに組織分布および補因子の使用の面で異なります。ヘキソキナーゼは、関連するグルコース代謝経路の大半において第一段階を触媒しています。ヘキソキナーゼ活性の変動は、X染色体性筋ジストロフィー、溶血性貧血、および癌など複数の疾患と関連しています。[2]

アプリケーション

比色法によるヘキソキナーゼ活性定量アッセイキットは、ヘキソキナーゼの活性を測定するために使用されています。[3][4][5]

適合性

ヘキソキナーゼ活性の測定、グルコース代謝および細胞シグナル伝達の分析、ならびに血清、動物組織（肝臓、心臓、腎臓など）および細胞培養（接着細胞または浮遊細胞）の糖尿病薬のスクリーニングに適しています。

原理

比色法によるヘキソキナーゼ活性定量アッセイキットは、様々な試料のヘキソキナーゼ活性を簡便かつ直接的に測定する手順を提供します。ヘキソキナーゼ活性は共役酵素反応により測定され、グルコースはヘキソキナーゼによりグルコース-6-リン酸に変換され、グルコース-6-リン酸デヒドロゲナーゼにより酸化され、NADHを生成します。生成されたNADHは無色のプローブを還元し、その結果、ヘキソキナーゼ活性の存在量に比例した比色分析（450 nm）可能な生成物が生じます。1単位のHKは、pH 8.0および室温で1分間に1.0 μモルのNADHを生成する酵素量に相当します。

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.
