Applicazioni

La soluzione di insulina da pancreas bovino è stata usata per studiare:
  • l'effetto di insulina esogena sulle concentrazioni plasmatiche di glucosio, insulina, fattore di crescita insulino-simile I (IGF-I) e progesterone (P4) in giovenche in età puberale[1]
  • la responsività all'insulina della placenta nel corso di complicanze gravidiche utilizzando trofoblasti umani primari ed espianti di villi placentari[2]
  • l'effetto della trasduzione del segnale del recettore insulinico sulla densità delle spine dendritiche di neuroni ippocampali neonatali murini in vitro[3]

Azioni biochim/fisiol

L'insulina è uno dei principali ormoni che regolano il glucosio nel sangue e la cui insufficienza causa l'iperglicemia caratteristica del diabete di tipo 2.[4]
Ormone polipeptidico a doppia catena prodotto dalle β-cellule delle isole pancreatiche. Il suo peso molecolare è circa 5800 Da. Le catene α e β sono unite da due legami disolfuro. La catena α contiene un legame disolfuro interno. L′insulina regola la captazione, l′utilizzo e lo stoccaggio di glucosio, aminoacidi e acidi grassi a livello cellulare e inibisce la disgregazione di glicogeno, proteine e grassi.

    1. The product has not been validated for rat cell culture application. However, based on the following citations, it appears to work well with rat cell culture:

      published in J Vis Exp. 2020 Oct 10;(164):10.3791/61073. PMID: 33104067.

  2. Does this solution contain zinc? How would that affect how quickly the insulin would act in a mammalian cell culture?

    1. The product contains 10 mg/mL insulin in 25 mM HEPES, pH 8.2, but it does not include zinc. Currently, the effect of zinc on insulin signaling is not characterized. However, it is noted that zinc appears to be able to enhance insulin-mediated IR tyrosine phosphorylation according to the article Biol Trace Elem Res. 2004 Oct;101(1):19-36. Effect of chromium and zinc on insulin signaling in skeletal muscle cells. PMID: 15516700.

  3. is this product suitable for 3T3-L1 differentiation?

    1. This product is supplied as a sterile bovine insulin solution at 10 mg/mL and would be suitable for diffentiation assays. This item has not been tested on 3T3-L1 cells, but is part of the differentiation protocol for the WT-1 preadipocyte cell line, product SCC255. See the link below to review this protocol:

    1. This solution is made using insulin Product No. I5500.  The potency specification for the I5500 is not less than 27 USP units/mg.  However, we do not test for potency of the final product of this solution.  It is possible some activity is lost during the manufacturing process.

    1. Product No. I0516 is a 10 mg/mL solution of bovine insulin (MW 5733.5). The molarity is 1.7 μmol/mL or 1.7 mM.

  8. What is the comparison between International Units (IU) and USP units for insulin?

    1. The international unit (IU) of insulin is based on the International Standard Preparation of Bioassay (1958). The standard preparation is a mixture of 52% bovine and 48% porcine insulin, containing 24 units/mg. Reference: Martindale: The Extra Pharmacopoeia, 29th ed., p. 392. High purity bovine pancreatic insulin typically assays at approximately 27 USP units per mg. So the IU and USP unit are not identical, but they are approximately 1:1.
