General description
L′albumina sierica bovina (BSA) è una proteina plasmatica ad α-elica, globulare, non glicosilata, prodotta dal fegato dei bovini. Consiste di tre domini (I, II e III), ognuno dei quali presenta due sub-domini (A e B), organizzati in una struttura a forma di cuore.
Application
L′albumina sierica bovina è stata impiegata:
- insieme alla BTS (soluzione da scongelamento di Betsville) per il prolungamento della vita degli spermatozoi durante l'analisi di motilità
- come componente del tampone Krebs-Ringer modificato (dmKRBT)
- in analisi di gel shift[1]
- come agente bloccante in immunocitochimica, immunoistochimica,(4) in protocolli di immunofluorescenza e nel saggio di immunoassorbimento enzimatico (ELISA) a cattura (ELISA a sandwich)
- come componente di una soluzione di tampone fosfato salino per risospendere le cellule da sottoporre ad analisi mediante citometria di flusso
- come standard proteico per la quantificazione di proteine in campioni tissutali con il metodo di Bradford
- per determinare l′attività dell′enzima 17β HDS

Si sospetta che alcuni epitopi conformazionali e di sequenza primaria di BSA siano allergeni nelle allergie umane a carne bovina e latte.
Preparation Note
Chiamato anche frazione di Cohn V, questo prodotto è ottenuto mediante una modifica del processo di Cohn per il frazionamento del plasma a freddo con etanolo.
Ci si riferisce alla sieroalbumina come Frazione V. Questa designazione è presa dal metodo originale introdotto da Cohn per frazionare siero proteine usando la precipitazione in etanolo a freddo. La sieroalbumina è stata trovata da Cohn nella quinta frazione col suo metodo basato sull′etanolo. Da allora il termine “Frazione V” è stato usato da alcuni a prescindere dal metodo di preparazione. Sigma-Aldrich produce e distribuisce sieroalbumine purificate con vari metodi primari inclusi il metodo originale di frazionamento di Cohn, metodi modificati di frazionamento con etanolo, shock termico e cromatografia. Stadi successivi di purificazione possono comprendere cristallizzazione o filtrazione su carbone attivo.
