Anwendung

SIGMAFAST™ OPD(o-Phenylendiamin-dihydrochlorid)-Tabletten wurden zur Verwendung als lösliches Substrat für den Nachweis einer Peroxidase-Aktivität in Enzym-Immunassays (EIA) entwickelt. OPD ist das EIA-Substrat erster Wahl, da es eine hohe Sensitivität aufweist. Die Ergebnisse von EIA-Tests, in denen OPD verwendet wird, können entweder nach einer bestimmten Zeit abgelesen oder mit verdünnter Säurelösung gestoppt werden, wenn die Ergebnisse zu einem späteren Zeitpunkt abgelesen werden sollen.
SIGMAFAST OPD wurde für VLP-Bindungsassays (virus-like particles) mit Speichelproben verwendet.[1] Es wurde auch zur Entwicklung von ELISA-Platten (Enzyme-linked Immunosorbent-Assay) zum Nachweis von Rituximab in Serumproben von Ratten verwendet.[2]

Leistungsmerkmale und Vorteile

SIGMAFAST™ OPD-Tabletten erfordern keine zusätzlichen Puffer oder Schritte für das Ansetzen einer aktiven Substratlösung.

Das Substrat ist hygroskopisch und lichtempfindlich

Rekonstituierung

Eine OPD-Tablette und eine Harnstoff-Wasserstoffperoxid-/Puffer-Tablette ergibt, in 20 ml Wasser gelöst, 20 ml gebrauchsfertiges Substrat. Wenn die Tablette auf diese Weise aufgelöst wird, betragen die Endkonzentrationen 0,4 mg/ml OPD, 0,4 mg/ml Harnstoff-Wasserstoffperoxid und 0,05 M Phosphat-Citrat, pH-Wert 5,0. Die Tabletten sind einzeln in Folienpackungen verpackt.
Jeder in 20 ml entionisiertem Wasser aufgelöste Tablettensatz ergibt eine gebrauchsfertige gepufferte Lösung, die OPD und Harnstoff-Wasserstoffperoxid enthält.

    1. Product P8287, o-Phenylenediamine dihydrochloride tablet will work, but it will require the addition of phosphate-citrate buffer and hydrogen peroxide.   2. Can I make the solution in large quantity and keep it in the fridge? Or should the solution be used right away because of its light sensitivity?

    1. The Substrate Solution is light-sensitive and should be protected from direct sunlight or UV sources in a
      tightly capped amber bottle. For best results, the Substrate Solution should be used within one hour.
