Die antithrombotische Wirksamkeit bei parenteraler Verabreichung kann durch Integration in die kationischen Liposome, die als eine Art von Retardanzien wirken, wesentlich verlängert werden.

Physikalische Eigenschaften

Das Antikoagulans Hirudin ist ein saures Protein mit einem Molekulargewicht von ~7 kDa und 65 Aminosäureresten. Es kommt in Blutegeln (Hirudo medicinalis) natürlich vor. Es ist der stärkste Inhibitor sowohl von löslichem (freiem) als auch gerinnselgebundenem Thrombin. Hirudin bindet Thrombin mit hoher Affinität und deckt mehr als 20 % der Thrombinoberfläche, wobei der aktive Situs und der exolytische Situs I (Fibrinogen und PAR-Erkennung) okkludiert werden. Durch diese umfassende Deckung werden Thrombuswachstum und Plättchenaktivierung blockiert. Hirudin wird in der menschlichen Blutbahn nicht metabolisiert und scheidet unverändert durch die Nierenfiltration aus. Natives Hirudin enthält einen sulfatierten Tyrosylrest (Tyr63), drei Disulfidbrücken und einen hohen Anteil an Dicarboxylsäuren. Hirudin wird nicht glykosyliert und enthält keine Tryptophan-, Arginin- und Methioninreste. Mindestens 20 Isoformen wurden identifiziert und sequenziert.
Dieses rekombinante Hirudinprodukt entspricht der HV-1-Variantensequenz, jedoch mit der Ausnahme, dass der Tyr63-Rest nicht sulfatiert wird.

Einheitendefinition

Gemäß einem direkten Vergleich mit einem NIH-Trombin-Referenzstandard neutralisiert eine Antithrombin-Einheit (ATU) eine NIH-Einheit Thrombin bei 37 °C.

Rekonstituierung

Hirudin ist wasserlöslich. In der Fachliteratur wird die Verwendung von „Verdünnungsflüssigkeit II“ (35,7 mM Essigsäure, 35,7 mM Natriumdiethylbarbiturat, 0,85 % NaCl, 1 % Rinderserumalbumin und 0,5 % PEG, pH 7,4) für die Auflösung von Hirudin (500 ATU/ml) und Thrombin beschrieben. Berichten zufolge ist Hirudin in Pyridin löslich, aber in Alkohol, Ether, Aceton oder Benzol praktisch unlöslich.

  1. Hi Please help me for reconstituting of Hirudin H0393  H100UN . Can I solve it by distilled water ? How much of DW should be added to the vial? Another question is the vial seems to be empty however it is plumb . Is this because the amount of powder?

    1. This material is soluble in water, a rate has not been established. For assay purposes, this product is reconstituted at 10 units/100ul in 28.6 mM Barbital buffer (see product B5934) containing 1.0% (w/v) Bovine Serum Albumin and 0.5% (w/v) Polyethylene Glycol, pH 7.35.       The activity specification for this product is minimum 7000 units per milligram protein. Occasionally, a small amount of powder may not be visible. A gentle spin or ‘shake down’ of the vial may move the powdered product to the bottom of the vial.

  2. Thanks for the previous answer, but the "units" definition is confusing. Does "100-units pack" mean 100-IU (in the International Standard system) or -ATU again?

    1. One antithrombin unit (ATU) will neutralize one NIH unit of thrombin at 37°C, based on a direct comparison to an NIH thrombin reference standard. There is no International Standard Unit value associated with this product.
