Allgemeine Beschreibung

Diphtherietoxin kann als Toxinelement bei der Konstruktion eines Immuntoxins für die zellspezifische Zytotoxizität verwendet werden. Die Translokationsdomäne der bakteriellen Toxine mit natürlichem Endosomen-Fluchtmechanismus wird für die Entwicklung effizienter nichtviraler Vektoren für Anwendungen in der Gentherapie verwendet.

Anwendung

Diphtherietoxin aus Corynebacterium diphtheriae wird wie folgt verwendet:
  • zur Depletion von Mikroglia, um posttraumatische Belastungsstörung bei Mäusen zu untersuchen[1]
  • als intraperitoneale Injektion in transgenen Mäusen, um die Hybride nach der Fusion von transplantierten hämatopoetischen Stamm- und Vorläuferzellen (HSPC) bei der Ablation zu selektieren[2]
  • zur Untersuchung seiner Wirkung auf die auf die Eosinophillinie festgelegten Vorläuferzellen in einem eosinophildefizienten Mäusestamm (iPHIL)[3]

Biochem./physiol. Wirkung

Diphtherietoxin ist ein bakterielles Toxin, das aus Corynebacterium diphtheriae erzeugt wird. Es enthält drei Domänen, die intrazelluläre Wirkungen aufweisen. Die Domänen sind an der Vergiftung der Zelle, der Zelloberflächenbindung und Internalisierung in Endosomen, der Translokation durch die Endosommembran in das Zytosol und die Hemmung der zellulären Proteinsynthese beteiligt. Diphtherietoxin ist nützlich zum Entwickeln von biotechnologischen Werkzeugen und Therapeutika.[4] Es hemmt die Proteinsynthese, indem es die Ribosylierung des Zielproteins durch Adenosindiphosphat (ADP) in Eukaryoten katalysiert.[5]

Physikalische Form

Lyophilisiertes Pulver, das Tris und EDTA enthält.

Rekonstituierung

Jede mit 0,5 ml sterilem destilliertem Wasser rekonstituierte Flasche enthält ~1 mg Diphtherietoxin in 0,01 M Tris und 0,001 M Na2EDTA, pH 7,5.

  2. What is recommend for reconstituting the product for injection in mice via ip injection? The product instructions advise using sterile distilled water, but the product also contains Tris and EDTA. Most references and research articles recommend using PBS for reconstitution. I am concerned that water might be too hypotonic for the mice and may cause their cells to rupture, but I'm not sure whether the Tris and EDTA can prevent that.

    1. No test has been conducted on this product for intraperitoneal injection. However, for intraperitoneal injection in mice, normal saline solution appears to be effective as a delivery solvent

MW 39,000 and 24,000 Daltons (corresponding to the B and A fragments, respectively). Based on this method, the extent of the degree of nicking in this preparation is estimated to be less than 5%.

    1. Once the product is reconstituted in water, the solution may be stored in aliquots at -20°C. Avoid freeze-thawing. Long-term storage in the refrigerator at 2-8°C is not recommended.
