Aplicação

Os polímeros poli-D-lisina podem ser usados para preparar superfícies para adesão celular. Os polímeros D-lisina também podem ser usados com células que digerem polímeros poli-L-lisina e causam captação excessiva de L-lisina.

Este produto é recomendado como substrato de cultura celular quando se usa 0,5-1,0 ml de uma solução de 0,1 mg/ml para cobrir 25 cm2. As versões de menor peso molecular do produto são menos viscosas, mas versões com maior peso molecular fornecem mais sítios de adesão por molécula.

Ações bioquímicas/fisiológicas

O bromidrato de poli-D-lisina (PDL) é um fator de fixação inespecífico para células, útil para promover a adesão de células a substratos sólidos, aumentando a interação eletrostática entre íons carregados negativamente da membrana celular e a superfície da cultura. Após absorção à superfície da cultura celular, a poli-D-lisina atua para aumentar o número de sítios de ligação celular com carga positiva.

Componentes

A poli-D-lisina é um polímero de aminoácido com carga positiva, com cerca de um HBr por resíduo de lisina. O hidrobrometo permite que a poli-D-lisina esteja em forma cristalina, solúvel em água. Uma pequena quantidade de produto pode ser encontrada na estrutura beta, pois o HBr interfere nas pontes de hidrogênio entre os grupos amino e carboxila, ou amino e as porções contendo N ou O.

Atenção

As soluções estéreis são estáveis por até 2 anos se armazenadas a 2 a 8 °C. Deve ser armazenado dessecado a -20 °C.

Nota de preparo

Para remover HBr, dissolva o produto em tampão neutro e faça a diálise para remover os sais. De modo geral, para usar este produto como fator de adesão, adicione 50 ml de água estéril para cultura de tecidos a 5 mg de polilisina e cubra assepticamente a superfície com 1 ml por 25 cm2 de solução. Após 5 minutos, remova a solução por aspiração e enxágue bem a superfície. Deixe secar por duas horas antes de introduzir as células e o meio.

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.
