Descrição geral

Agarose is a polymer extracted from agar or agar-bearing marine algae. This purified linear galactan hydrocolloid comprises alternating co-polymers D-galactose and 3,6-anhydro-L-galactose units connected by α-(1→3) and β-(1→4) glycosidic bonds.[1] Agarose is highly biocompatible and possesses variable mechanical and diffusion properties.[2] Agarose can be used as a gelling agent, to separate nucleic acids electrophoretically. Low melting or low gelling temperature agarose is produced by hydroxyethylation of agarose. It is usually used for the isolation of separated DNA fragments. DNA fragments of the equal size will take longer time to move through a low melting agarose gel compared to a standard agarose gel.[3]

Aplicação

Gels exhibit excellent clarity and are particularly useful for the preparation of media containing heat-labile materials. Recommended for preparation of agarose beads.
Agarose, low gelling temperature has been used:
  • to immobilize zebra fish embryos during in vivo imaging experiments[4]
  • as a constituent of defined medium to culture bovine nucleus pulposus (NP) cells[5]
  • as a hydrogel, a scaffolding material for dental pulp regeneration[6]
  • in the form of pellets to determine the surface pH and conductivity of an acrylic emulsion film[7]

Teor de sulfato - usado como indicador de pureza, uma vez que o sulfato é o principal grupo iônico presente.
Força do gel - a força que deve ser aplicada a um gel para que se frature.
Ponto de gel - a temperatura na qual uma solução de agarose aquosa forma um gel ao se resfriar. Soluções de agarose exibem histerese na transição líquido para gel, ou seja, o seu ponto de gel não é o mesmo que a sua temperatura de fusão.
Eletroendosmose (EEO) - um movimento de líquido através do gel. Grupos aniônicos em um gel de agarose são afixados à matriz e não podem se mexer, mas contracátions dissociáveis podem migrar no sentido do cátodo na matriz, o que dá origem à EEO. Como o movimento eletroforético de biopolímeros é, em geral, no sentido do ânodo, a EEO pode perturbar separações em função da convecção interna.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   3. In order to have sterile Low Melting Agarose, once the package has already been opened outside of sterile conditionds, should I filter it and then lyophilize or should I use an autoclave?
