Description générale

PNMT, also known as Phosphatidylethanolamine N-methyltransferase, or PEMT, PEAMT, or PEMT2, and encoded by the gene PEMT/PEMPT/PNMT, catalyzes three sequential methylation reactions of phosphatidylethanolamine to produce phosphatidylcholine a major constituent of cell membranes and a primary pulmonary surfactant. PNMT/PEMT is localized to the endoplasmic reticulum membrane as well as the mitochondrial membranes. PNMT/PEMT is widely expressed, and has the highest activity in the mitochondria. The conversion of phosphatidylethanolamine to phosphatidylcholine by sequential methylation via PNMT/PEMT occurs in the liver. A different pathway for phosphatidylcholine production starting from choline also is present in cells but it does not involve PNMT/PEMT enzyme. Mutations in PNMT/PEMT protein are associated with nonalcoholic fatty liver disease (NAFLD).

Immunogène

Epitope: C-terminus
KLH-conjugated linear peptide corresponding to the C-terminus of human PNMT.

Application

Anti-PNMT Antibody detects level of PNMT & has been published & validated for use in PNMT.
Immunohistochemistry Analysis: A 1:500 dilution and a 1:2,000 dilution from a representative lot detected PNMT in human adrenal gland and testis tissues, respectively.

Qualité

Evaluated by Western Blotting in human liver tissue lysate.

Western Blotting Analysis: 2 µg/mL of this antibody detected PNMT in 10 µg of human liver tissue lysate.

Description de la cible

~17 kDa observed. As isoform of PNMT has been observed between 15-20 kDa (Cui, Z., et al. (1993). J Biol. Chem. 268(22):16655-16663.).
