General description
Protamine sind Proteine mit hohem Anteil an Cystein und Arginin. Sie besitzen viele Phosphorylierungsstellen. Sie befinden sich im Spermienkopf.
Application
Protaminsulfatsalz aus Lachs wird wie folgt verwendet:
- bei der Vorbehandlung von Zellen zur Untersuchung seiner Wirkung auf eine Infektion mit respiratorischem Synzytial-Virus
- bei der Herstellung von Lentivirus
- zur Derivatisierung in einen Diazeniumdiolat-NO-Donator
Kann zum Entfernen von DNA aus Proteinproben und zur Aufreinigung von DNA-bindenden Proteinen eingesetzt werden. Es kann als Alternative zu Polybren beim Retroviren-vermittelten Gentransfer eingesetzt werden.
Protamin in Form von festen Lipidnanoteilchen (SLN) fördern die Transfektion mit Plasmid-DNA wirksamer und mit weniger Zytotoxizität als vergleichbare SLN aus Esterquat-1.

Biochem/physiol Actions
Protaminsulfat ist ein kleines kationisches Protein, das DNA bindet und ausfällt. Neutralisiert die gerinnungshemmende Wirkung von Heparin. Hemmt Lipoproteinlipase.
