Allgemeine Beschreibung

Dieser Proteaseinhibitor-Cocktail wurde für den allgemeinen Gebrauch optimiert und geprüft. Er besteht aus wasserlöslichen Proteaseinhibitoren, die Serin-, Cystein- und Metalloproteasen wirksam hemmen. Dieses Produkt wird als lyophilisiertes Pulver bereitgestellt.

Spezifität

Hemmt Serin-, Cystein- und Metallproteasen.

Anwendung

Proteaseinhibitor-Cocktail-Pulver wird als Komponente verwendet:
  • in Puffer für Radioimmunopräzipitationsassays zur Lyse von Kristallinproben der Augenlinse von Vögeln für die Western-Blot-Analyse[1]
  • in Phosphatpufferlösung (PBS) zum Mahlen von Blättern, die rekombinante Proteine exprimieren, um virusähnliche Partikel (VLPs) aufzureinigen[2]
  • in PBS zur Auflösung einer lyophilisierten Hautsekretprobe von A. loloensis zur Peptidaufreinigung[3]
  • in 0,1 M NaCl-Lösung, um Hautsekrete von Fröschen zu waschen und zu sammeln[3]

Biochem./physiol. Wirkung

Die Bestandteile dieser Mischung wie AEBSF und Aprotinin hemmen Serinproteasen, unter anderem Trypsin, Chymotrypsin und Plasmin. Bestatin hemmt Aminopeptidasen. E-64 wirkt gegen Cysteinproteasen. EDTA hemmt Metallproteasen. Leupeptin wirkt sowohl gegen Serin- als auch Cysteinproteasen.

Komponenten

Diese Mischung enthält folgende Bestandteile:

2 mM AEBSF
0,3 μM Aprotinin
116 μM Bestatin
14 μM E-64
1 mM EDTA
1 μM Leupeptin

Jede Komponente hat spezifische hemmende Eigenschaften.

Vorsicht

Das lyophilisierte Pulver ist bei einer Lagertemperatur von -20 °C 4 Jahre lang haltbar.

Menge

Eine Flasche ergibt 100 mL des Cocktails, eine Menge, die zur Hemmung der Proteasen in ca. 20 g Zellmasse ausreicht.

Nicht alle Extrakte enthalten dieselbe Zahl endogener Enzyme und ggf. muss die Menge des benötigten Cocktails angepasst werden.

Angaben zur Herstellung

Dieses Produkt wird als lyophilisiertes Pulver bereitgestellt. Eine Flasche ermöglicht die Zubereitung von 100 mL einer Cocktaillösung.

    1. P2714 includes EDTA at 1 mM. The components of this mixture, such as AEBSF and aprotinin, act to inhibit serine proteases, including trypsin, chymotrypsin, and plasmin among others. Bestatin inhibits aminpeptidases. E-64 acts against cystein proteases. EDTA is an inhibitor of metalloproteases. Leupeptin acts against both serine and cystein proteases.

  3. Wie löse ich das Pulver in 100 ml aqua dest???? Wie lagere ich das dann?

    1. This product is packaged in a 10 mL serum vial with a crimp cap. Dissolve the contents of the vial in 10 mL of water or working buffer, then transfer to another container for dilution to 100 mL. Store the solution in working aliquots for at least 1 month -20 C, to avoid repeated freeze/thaw cycles. See the link below to review the product datasheet:

  4. I intend on taking 1mL samples of blood to give serum and test hormone levels in these samples. Should I add the solubilised protease inhibitor cocktail (p2714) directly to the blood or after centrifugation (which could be a few hours later)?

    1. A short delay may be negligible, however, it would not be detrimental to add protease inhibitors to whole blood samples if an extended delay is anticipated. This would ensure the best results. Please see the link below to review a publication of a similar nature:

    1. The amount in each vial is 89 mg. This material is very hygroscopic. It is recommended to prepare the entire package rather than attempt to weigh out specific amounts.

    1. You can dissolve the product in 10 ml of water to make a 10X stock solution and then dilute 1:10 for use.  Aliquots can be stored at -20 C for 1 month.
