Application
Zur Verwendung als lösliches Substrat für den Nachweis einer alkalischen Phosphatase-Aktivität in Enzym-Immunassays (EIA) entwickelt. pNPP ist das bevorzugte Substrat für hochempfindliche alkalische Phosphatase-basierte EIA-Assays. Eine pNPP-Tablette und eine Tris-Puffer-Tablette werden in entionisiertem Wasser aufgelöst, wodurch ein gebrauchsfertiges Substrat gewonnen wird. Jeder SIGMAFAST-pNPP-Tablettensatz liefert eine Lösung mit 1,0 mg/ml pNPP und 0,2 M Tris-Puffer. Die Ergebnisse von EIA-Tests, in denen pNPP verwendet wird, können entweder nach einer bestimmten Zeit abgelesen oder mit 3 N NaOH gestoppt werden, wenn die Ergebnisse zu einem späteren Zeitpunkt abgelesen werden sollen. Die SIGMAFAST™-Tabletten benötigen zum Ansatz einer aktiven Substratlösung lediglich die Zugabe von destilliertem oder entionisiertem Wasser. Die Tabletten sind einzeln in Folienpackungen verpackt.
Preparation Note
Jeder in 20 mL entionisiertem Wasser aufgelöste Tablettensatz ergibt eine gebrauchsfertige gepufferte Lösung, die pNPP enthält.
