Allgemeine Beschreibung

Citrat ist ein wichtiges Tricarbonsäure(TCA)-Zyklus-Zwischenprodukt, das durch die Zugabe von Oxalessigsäure zur Acetylgruppe von Acetyl-CoA entsteht. Citrat wird über das Citrat-Malat-Shuttle aus den Mitochondrien heraus transportiert und für die Fettsäuresynthese wieder zu Acetyl-CoA umgewandelt. Citrat ist ein allosterischer Modulator der Fettsäuresynthese infolge seiner Wirkung auf die Acetyl-CoA-Carboxylase und der Glykolyse infolge seiner Wirkung auf die Phosphofructokinase. Citratstoffwechsel und -disposition können sich je nach Geschlecht, Alter und einer Vielzahl anderer Faktoren (auch Krankheitsstadien) deutlich unterscheiden. Der Citratspiegel in Zellen wird in Prostatakrebszellen reduziert; der Citratspiegel kann ein Marker für die Wachstumsrate von Prostatakrebs sein. Das Citrat-Assay-Kit bietet ein einfaches, empfindliches und schnelles Verfahren zur Quantifizierung von Citrat in verschiedenen Proben.

Anwendung

Das Citrat-Assay-Kit wird für die Bestimmung der Citratkonzentration in Proben verwendet.[1][2][3]

Eignung

Für das Messen von Citrat in vielen verschiedenen Proben, einschließlich Gewebe und Zellen, geeignet.

Prinzip

Die Citratkonzentration wird durch ein gekoppeltes Enzym-Assay bestimmt, das – proportional zum vorhandenen Citrat – ein kolometrisches (570 nm)/ fluorometrisches (λex = 535/λem = 587 nm) Produkt ergibt. Die typische Empfindlichkeit des Citrat-Assay-Kits liegt zwischen 0,1 und 10 nmol (2 μM-10 mM) Citrat in verschiedenen Proben.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   3. How are the detection range and sensitivity determined for the MAK057 assay?

    1. The calculation of the higher limit of detection is determined by dividing the highest concentration of the standard by the minimum sample volume. Similarly, the lower limit of detection is calculated by dividing the lowest concentration standard by the maximum sample volume. In the event that a sample exceeds the range of the standard curve, it is advisable to conduct a dilution and then adjust for the dilution factor.
      For instance, for the MAK057 test:

      Colorimetric:
      • Lower range: 2 nmol/50 = 0.04 mM or 40 uM
      • Higher range: 10 nmol/1 uL = 10 mM or 10,000 uM
      Fluorometric:
      • Lower range: 0.2 nmol/50 = 0.004 mM or 4 uM
      • Higher range: 1 nmole/1 uL = 1 mM or 1000 uM
