General description

Recombinant human Interleukin‑6 (IL‑6) is a cytokine expressed in HEK 293 cells and supplied as a lyophilized protein. IL‑6 is commonly used in mammalian cell culture research involving immune‑related cell models.

Application

Recombinant IL‑6 has been used to elucidate its role in breast cancer metastasis and in D5HS tumor sphere formation.

Biochem/physiol Actions

Interleukin-6 (IL-6) ist ein multifunktionelles Protein, das ursprünglich in den Medien von Zellen entdeckt wurde, die mit doppelsträngiger RNA stimuliert wurden. IL-6 scheint direkt an den Reaktionen beteiligt zu sein, die nach einer Infektion oder Verletzung auftreten, und könnte sich in Bezug auf die Regulierung der Akute-Phase-Reaktion als ebenso wichtig wie IL-1 und TNF-α erweisen. Berichten zufolge wird IL-6 von Fibroblasten, aktivierten T-Zellen, aktivierten Monozyten oder Makrophagen sowie Endothelzellen produziert. Es wirkt auf eine Vielzahl von Zellen, darunter Fibroblasten, myeloide Vorläuferzellen, T-Zellen, B-Zellen und Hepatozyten. IL-6 induziert multiple Wirkungen, auf die seine zahlreichen Synonyme schließen lassen: Plasmozytom-Wachstumsfaktor (PCT-GF), Interferon-β-2 (IFN-β₂), von Monozyten abstammender humaner B-Zell-Wachstumsfaktor, B-Zell-stimulierender Faktor (BSF-2), Leberzell-stimulierender Faktor (HSF), Interleukin-Hybridom/Plasmozytom-1 (IL-HP1). Außerdem scheint IL-6 für die Proliferation von T-Lymphozyten mit IL-2 zu interagieren. IL-6 potenziert auch die proliferative Wirkung von IL-3 auf multipotente hämatopoietische Vorläuferzellen.

Interleukin-6 (IL-6) is produced by fibroblasts, activated T cells, activated monocytes or macrophages, and endothelial cells. It induces B-cell differentiation and is a primary regulator of inflammation. IL6 regulates tumor proliferation and inhibits apoptosis. It also potentiates the proliferative effect of IL-3 on multipotential hematopoietic progenitors.

Preparation Note

Human IL-6, expressed in human 293 cells, is a glycosylated monomer with an apparent molecular mass of 26-30 kDa due to glycosylation, which is absent when this cytokine is expressed in E. coli. Production in human HEK 293 cells offers authentic glycosylation. Glycosylation contributes to stability in cell growth media and other applications.

Analysis Note

The specific activity was determined by the dose-dependent stimulation of the proliferation of human TF-1 cells (human erythroleukemic indicator cell line).
