General description

Hyaluronidase ist in der Natur weit verbreitet. Im Menschen ist sie in einer Reihe von Organen und Körperflüssigkeiten vorhanden. Hyaluronidase ist in den Sekretionen von Viren, Bakteriophagen, Pilzen und Bakterien enthalten. Dies gilt auch für die Sekretionen von Nematoden und Blutegeln. Schlangen und Skorpione produzieren Hyaluronidase in ihrem Gift. Außerdem wird Hyaluronidase von Bienen, Spinnen, Wespen, Hornissen, Raupen, Fischen und Eidechsen abgesondert.

Application

Hyaluronidase von Streptomyces hyalurolyticus hat die folgenden Anwendungen:
  • zur Herstellung von DMEM/F-12-Medien (Dulbeccos modifiziertes Eagle-Medium/Nährstoffmischung F-12) für die Isolierung und Aufreinigung einzelner Zellen aus abgelöstem Tumorgewebe
  • als Bestandteil einer Verdaulösung für die Derivation von Keimbahn-Stammzellen aus testikulärem Gewebe

Biochem/physiol Actions

Hyaluronat-Lyase spaltet Hyaluronsäure an der β-D-GlcNAc-(1→4)-β-D-GlcA-Bindung ab, wodurch Tetra- und Hexasaccharide von 3-(4-Deoxy-β-D-gluc-4-enuronosyl)-N-acetyl-D-Glucosamin entstehen. Anders als andere Hyaluronidasen ist dieses Enzym spezifisch für Hyaluronsäure und mit Chondroitin und Chondroitin-Sulfaten inaktiv.
Hyaluronidase ist eine Endoglycosidase und führt ihre Funktion zusammen mit Exoglycosidasen wie β-Glucuronidase und β-N-Acetyl-Glucosaminidase durch. Hyaluronidase reguliert den Hyaluronsäureumsatz.

Other Notes

Eine Einheit baut 0,75 μg des Polysaccharids Hyaluronsäure pro Minute bei einem pH-Wert von 5,35 und einer Temperatur von 37 ºC ab (bestimmt durch turbidimetrische Messung der Absorption (λ: 600 nm), wenn sie nach 45 Minuten mit BSA komplexiert wird).

  1. What is the volume in each vial/ampoule?

    1. H1136 is not supplied in solution form. The product is lyophilized directly into the ampule and its appearance is a clear, thin film.

  2. You mention the following buffer composition for reconstitution: "...20 mM sodium phosphate buffer, with 77 mM sodium chloride, 0.01% BSA, pH 7.0". If I want to refreeze aliquots, would you suggest the same buffer? Isn't sodium phosphate harmful here?

    1. Long-term solution stability studies have not been performed on the microbial source hyaluronidase. However, the bovine source products are stable for up to 1 month at 4 deg. C (500 mM acetate buffer pH 6.0) and several months at - 20 deg. C. Aliquot solutions to store frozen, avoid repeated freeze/thaw cycles. The recommended reconstitution buffer should include 0.01% BSA which acts as a stabilizer, there is no indication that the recommended sodium phosphate/sodium chloride buffer would be inappropriate for freezing.
