After opening, this product is stable for at least 1 week at room temperature under inert gas such as nitrogen or argon. The long term storage of this product has not been determined. However, the powder material, under product number 580560, reports that TCEP may be stored at -20°C in aliquots for up to 3 months. Note that this material in solution retains stability when stored at an acidic or alkaline pH. Adjustment of the pH would be required prior storage. Avoid storage at pH 7.0-8.0. Also avoid exposure to high concentrations of phosphate ions (>150 mM).

Das Produkt gehört zur Klasse der Trialkylphosphine.[1]
Tris (2-Carboxyethyl)-Phosphin (TCEP) ist sehr wirksam beim Aufspalten von Disulfidbindungen in wässrigen Lösungen.[2] Im Gegensatz zu anderen Trialkylphosphinen (Tributylphosphin) ist es wasserlöslich und geruchlos. Es ist auch weniger toxisch als 2-Mercaptoethanol. Aufgrund dieser Vorteile ist das Produkt besser als andere Reduktionsmittel.[3]
Anwendung
Tris (2-Carboxyethyl)-Phosphin (TCEP) kann bei verschiedenen nachgelagerten Anwendungen eingesetzt werden, einschließlich SDS-PAGE, Massenspektrometrie, Kennzeichnung mit cysteinspezifischen Markierungen und Modifikation von cysteinhaltigen Verbindungen. Das Produkt verhindert die Oxidation von Proteinproben. Daher ist es eine nützliche Pufferkomponente, da es die Enzymaktivität erhält.[4][5][6] Es wird in der Reduktion und Messung von Glutathion verwendet.[7]
Tris (2-Carboxyethyl)-Phosphin (TCEP) wird auch für Folgendes verwendet:
- zum Spalten von Cysteinresten in einem synthetischen Peptid[5]
- in einem Reduktionspuffer für RNA Sequentielle Untersuchung der Targets (SPOTs)-Bildgebung[8]
- zur Reduktion von Oligonukleotiden[9]
- als Reduktionsmittel bei der mitochondrialen Isolierung[10]
Biochem./physiol. Wirkung
Als Reduktionsmittel, das kein Mercaptan enthält, wird die in Thiol enthaltenden Verbindungen vorliegende Toxizität ausgeschlossen. Das Produkt ist in der Lage, den Schwerketten-/Leichtketten-Komplex Botulinum-Neurotoxin B aufzuschließen, der von einer einzelnen Disulfidbindung zusammengehalten wird und für die Endozytose sowie letzten Endes die Toxizität des Giftstoffs verantwortlich ist. Da disulfidgekoppelte Untereinheiten typisch für viele Giftstoffe sind (z. B. Rizin, Schlangengift und alle BoNT-Serotypen) ist das Produkt möglicherweise als Rescue-Prophylaktikum in Fällen der Verabreichung von Giftstoffen hilfreich.[11]
Tris(2-Carboxyethyl)-Phosphin-Hydrochlorid-Lösung reduziert die Disulfidbindungen und lässt andere funktionale Gruppen in Proteinen unberührt.[12]

How can I determine the shelf life / expiration / retest date of this product?

If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice.
