MAB80138 anti hMPV antibody was used in Immunofluorescence in the following study. A549 or WI-38 cells were fixed in PBS containing 4% paraformaldehyde for 10 minutes on ice, followed by permeabilization with 0.1% saponin for 30 minutes and blocking with PBS containing 10% fetal bovine serum and 2.5% bovine serum albumin for 60 minutes at room temperature. The primary antibody against hMPV nucleoprotein (N protein) was incubated overnight at 4°C, followed by secondary antibody GAM 647 for 30 minutes at room temperature. Allgemeine Beschreibung
Das humane Metapneumovirus (hMPV) gehört zur Pneumovirinae-Unterfamilie der Paramyxoviren, die erstmals 2001 in den Niederlanden anhand von pädiatrischen Atemwegsproben beschrieben wurden. Es handelt sich um ein umhülltes pleomorphes Virus mit einer einzelnen Minusstrang-RNA-Genom. Zwei Hauptserotypen wurden beschrieben: A und B. Das hMPV wurde in mehreren Studien weltweit in Proben nachgewiesen. Schätzungen zufolge haben 70 % der Kinder im Alter von 5 Jahren Antikörper gegen hMPV.
Spezifität
Erkennt das Nukleoprotein. Reagiert mit allen hMPV-Subtypen: A1, A2, B1 und B2.
Anwendung
Nachweis des humanen Metapneumovirus mittels dieses Anti-humanes-Metapneumovirus-Antikörpers (Klon 507), der für die Verwendung in ELISA, FC, IF und WB validiert wurde.
