应用

溴化乙锭 (EtBr) 是 PAGE 或琼脂糖凝胶电泳最常用的核酸染色剂。EtBr 的荧光在与双链 RNA 结合后可增加 21 倍,在与双链 DNA 结合后增加 25 倍,因此在低染色浓度(10μg/ml)下无需本底脱色。溴化乙锭已用于大量核酸荧光测定。 它已被证明可与单链 DNA(虽然没有那么强)和三链 DNA 结合。由于它能与 DNA 结合,EtBr 是 DNA 聚合酶的抑制剂。

生化/生理作用

溴化乙啶可插入双链 DNA 和 RNA 中,并作为移码诱变剂。它还可与吖啶橙结合用于区分存活的、凋亡的和坏死的细胞。

重悬

对于在电泳后进行凝胶染色,应先用水将储存液样品稀释到 0.5μg/ml,再将凝胶孵育 15-30 分钟。通常无需脱色,但是如果必须降低背景色,则可在水中脱色 15 分钟。然后可在紫外灯箱(254nm 波长)中检测 DNA 条带。也可将溴化乙啶加到凝胶和电泳缓冲液(终浓度 0.5μg/ml)中,并在电泳后立即显影。

If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.     1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

    1. Ethidium bromide is soluble in water at 100 mg/mL. Per Crary, B. and Boyrsenko, M., Biotechniques, 4, 98-101 (1986), stock solutions of ethidium bromide in water or PBS are stable for at least two years at room temperature if protected from light.

    1. For staining a gel after electrophoresis, dilute a sample of the stock solution to 0.5 μg/mL with water and incubate the gel for 15-30 min.  Ethidium bromide can also be incorporated into the gel and running buffer at 0.5 μg/mL, and visualized immediately after electrophoresis.

    1. Ethidium bromide is mutagenic, and ultimately has to be destroyed by incineration. It can be removed from buffer solutions by several methods to reduce the volume of waste. See the product sheet for more details.
